Red bulb plant or “bawang dayak” (Eleutherine americana Merr.) is commonly used as a traditional medicine for local Kalimantan people. This study was conducted to determine the genetic identity of red bulb plant species based on chromosomal character. The chromosomal study was carried out using a modified squash method. Mitosis period of E. americana mainly occurred from 10.00 to 11.30 a.m. (WITA-Center Indonesia Time). Prometaphase stages of red bulb plant species was commonly found at 10.30 a.m. (WITA). The species displayed chromosome numbers of 2n=12 (diploid) and the karyotype of 11 metacentric and 1 achrocentric (11M+1A) chromosomes with no satellite found in this study. The chromosomes of the complement in E. americana exhibited total length from 1.006 µm to 4.340 µm. This finding was indicated that chromosomes of E. americana species was heteromorphic, and the next study are necessary to be conducted.
