Coptosapelta tomentosa’s root empirically used as an antimalarial by people in Sungai Buah, Kotabaru, South Borneo. C. tomentosa’s root contains a variety of chemical compounds, the presence of chemical compounds which are non-polar such as anthraquinone, flavonoids and terpenoids can provide antimalarial activity. Non-polar chemical compounds separated from other chemical compounds using the fractionation method with n-hexane and aquadestillata. Antiplasmodial activity in the n-hexane fraction and water soluble fraction of C. tomentosa’s root was tested in vitro by inhibition of heme polymerization methods and measured with IC50 values. Chloroquine diphosphate was used as a positive control. DMSO 10% and aquades were used as a negative control. A significant difference between the IC50 value of the test compound and the positive control was analized by independent sample t-test with 95% trust level. The results showed that the inhibition percentage of heme polymerization from n-hexane fraction of C. tomentosa’s root with a concentration of 5; 2.5; 1.25; 0.625; 0.3125 mg/mL consecutively is 98.54; 94.60; 92.41; 85.87; 63.69%. IC50 value of heme polymerization test compound is 0,15 ± 0.01 mg/mL, water soluble fraction is fraksi 1.970 ± 1.38 mg/mL, and chloroquine diphosphate is 4.02 ± 0.63 mg/mL. The results of analysis with independent sample t-test declare that there is no difference between both of them with a significance value is 0.073. It can be concluded that n-hexane fraction of C. tomentosa’s root has an equal potential as good as chloroquine to inhibit 50% heme polymerization. |
